Effect of Platycodon Grandiflorum on the Blood Concentration of Carbamazepine in Rabbits

Abstract

OBJECTIVE: To study the interaction between traditional Chinese medicine and western medicine with a focus on the effect of Platycodon grandiflorum on the blood concentration of carbamazepine in rabbits.METHODS: After administration of carbamazepin alone or in combination with Platycodon grandiflorum(Platycodon grandiflorum was given for 1 time versus 7 times),the blood concentration of carbamazepine was determined with HPLC,with the blood concentration-time curve described and a statistical analysis conducted.RESULTS: The blood concentration of carbamazepine and the area under the curve were significantly higher in rabbits treated with carbamazepin in combination with Platycodon grandiflorum than those treated with carbamazepine alone.The blood concentration of carbamazepine at 2,4,and 6 hours were significantly higher in rabbits receiving Platycodon grandiflorum for 7 times than for 1 time(P0.05).CONCLUSION: The Platycodon grandiflorum can increase the blood concentration of carbamazepine,thus the dosage of carbamazepine could be reduced if used concomitantly with Platycodon grandiflorum so as to reduce incidence of ADR while without decreasing of its efficacy.This study results is of great importance as guidance for the clinical safe and rational drug use.
