Effect of Amlodipine on MCP-1 mRNA Expression in hUVEC-12 Induced by ox-LDL

Abstract

Objective To investigate the effects of amlodipine on the mRNA expression of monocytes chemotactic protein-1(MCP-1) in human umbilical vein endothelial cell(hUVEC) by oxidized low density lipoprotein(ox-LDL),so as to approach the mechanism of amlodipine in atherosclerosis. Methods hUVEC-12 were treated with ox-LDL in different concentration or different time.Based on that,the cells were pretreated with amlodipine at different concentrations(0,0.1,1.0,10.0 μmol/L) and then incubated with ox-LDL,MCP-1 mRNA expression was detected by reverse transcription-polymerase chain reaction(RT-PCR). Results MCP-1 mRNA expression induced by ox-LDL was obviously down-regulated by amlodipine in a dose-dependent manner. Conclusion Amlodipine may down-regulate the mRNA expression of MCP-1 induced by ox-LDL in hUVEC-12.
