Abstract
Objective To set up a stabilized and reliable model of rat permanent middle cerebral artery occlusion (pMCAO). Methods Rat pMCAO experiment was performed based on the previous Koizumi and Zea Longa's models with significant modification. Forty male Sprague-Dawley (SD) rats were randomly divided into experimental group and sham-operated group (n=20 in each group),and occurrence of rat cerebral ischemia in each group was assessed. In this study,2,3,5-Triphenyltetrazolium chloride(TTC) staining were carried out to detect the infarct area in the brain of experimental rats. Results In experimental group,the successful ratio of cerebral ischemia model was 95%,and neurological deficit scores of 1~3 grade was 95%. Conclusion This modified pMCAO model is an ideal model with better stabilization,higher reproducibility,easier execution and simpler operation when compared with the previous one.
