Abstract
Objective To determine the concentration of fentanyl.in human pLasma by HPLC with ult raviolet detection. Methods fentanyl was separated on Zorbax-CN coLumn with a mobile phase of water(containing 0.015 moL/L Na2HPO4 and 0.05 mol/L Pentane Sulfonic Acid Sodium Salt)-acetonitrile(70:30) and detected at 200 nm.Internal standard was remifentanil. The detection wavelength was 200nm.Results Calibration curve for fentanyl was linear in the range of 1~100ng/mL(r=0.999).The detection limit was 1.0ng/mL.The within-day RSD was(6.23±2.90) %,the between-day RSD was(6.45±4.12) %.The rate of the method recovery was(91.45±4.55) %.The rate of the extraction recovery was(95.63±4.02) %.Conclusion This method was quick and accurate for the determination of fentanyl in human plasma and useful for pharmakinetic study of fentanyl.
