Effect of penehyclidine hydrochloride on OA induced rat acute lung injury:a comparison between different administration methods

Abstract
Aim To investigate the therapeutic effect of penehyclidine hydrochloride in rats with oleic acid(0A)induced acute lung injury(ALI),and compare the effects between inhalation and intravenous therapy.Methods 30 male Sprague-Dawley rats were randomly divided into 3 equal groups:ALI group,penehyclidine hydrochloride inhalation group,and intravenous penehyclidine hydrochloride group.Blood pressure(BP),pulmonary artery pressure(PAP),artery blood gas analysis and mixed venous blood gas analysis were determined every 60 minutes during the experiment.The neutrophils and total protein(TP)content in bronchoalveolar lavage fluid(BALF),lung myeloperoxidase(MPO) activity,and the serum tumor necrosis factor-alpha(TNF-α) concentration were determined after the experiments in each group.Results The blood pressure was lower in intravenous group than that in other 2 groups.PAP,Paw and PaO2/FiO2 were improved in inhalation group and intravenous group,compared with ALI group.TP and neutrophils account in BALF,lung MPO activity and serum TNF-α concentration was lower in inhalation group and intravenous group than those in ALI group.Most parameters above are even better in inhalation group.Conclusion The improvement in the ALI model brought by inhaled penehyclidine hydrochloride is more effective than intravenous way.
