Abstract

Objective To study the content of functional composition of Semen Ziziphi Spinosae and its effect of improving sleep.MethodsThe content of coarse amylose,total flavone and total jujuboside were measured by chemical processes.The content of jujubosideA was measured by HPLC.The effect of sleep improvement of the three dosages(570 mg/kg,1000 mg/kg and 2000 mg/kg)of Semen Ziziphi Spinosae was studied by the experiments of sleep-induced directly,prolonging sleeping time induced by pentobarbital sodium,affecting mouse sleeping rate under the threshold hypnogenesis dosage of pentobarbital sodium and affecting sleeping delitescence induced by tobarbital sodium.The content of 5-HT in the brain of mouse was measured by fluorescence assay.ResultsThe content of total jujuboside in Semen Ziziphi Spinosae was 0.96 g/100 g,jujubosideA was 0.41 g/100 g,and total flavone was 0.62 g/100 g.The middle dosage(1 000 mg/kg) could improve the sleep quality,and sleep improvement had high relativity with 5-HT.ConclusionSleep can be improved by Semen Ziziphi Spinosae,and it has relation with the high content of the functional composition and the content of 5-HT in the brain of mouse.
