Optimization of microwave extraction technics of polysaccharides from Ganaderma lucidum and its model

Abstract

To optimize the extraction conditions of microwave technology of polysaccharides from Ganaderma lucidum by response surface methodology(RSM).The extraction parameters as ethanol concentration,extraction time,extraction power,the ratio of water to sample and the temperature of extraction were optimized based on single-factor experiment and three-level four-factor Box-Behnken design.It's regression coefficient was as high as 0.964 8.The optimum process conditions were obtained as follow: extraction temperature 90 ℃,extraction power 400 W ratio of water to sample 20 mL/g,extraction time 20 min.The average extraction rate of polysaccharides from Ganoderma lucidum was 1.150% with high stability.Second-order quadratic model is rather suitable.And compared with non-microwave extraction,the exaction rate of polysaccharides was increased significantly by microwave technology.It was concluded that microwave extraction had several remarkable advantages with shorter extraction time,less solvent and higher extraction rate.
