Determination of Paeonol and Paeoniflorin in Cortex Moutan from Various Habitats by HPLC

Abstract

A HPLC method was established for the determination of paeonol and paeoniflorin in cortex moutan from different habitats.The conditions of the chromatograph method was as following:A C18 column was used.The mobile phase for paeonol determination was methanol-water(50∶50) and detection wavelength was 274 nm.The mobile phase for paeoniflorin determination was acetonitrile-0.1%H3PO4(18∶82) and detection wavelength was 230 nm.The results show that the linear ranges of paeonol and paeoniflorin determination were 0.36~2.2 μg·L-1 and 0.042~1.06 μg·L-1 respectively.The average recoveries of paeonol and paeoniflorin were 96.30%(RSD=0.97%) and 98.21%(RSD=0.64%) respectively.The method is simple,accurate and reproducibility.The experimental results show that the contents of paeonol and paeoniflorin in cortex moutan from different habitats are positive relevant,while the contents of cortex moutan from Sichuan were higher than the others.
