Determination of Resveratrol in Polygonum cuspidatum by HPLC

Abstract

The contents of resveratrol in different growing condition and different part of Polygonum cuspidatum were detected by HPLC.The separation was performed on Zorbax CB-C18 column(250 mm×4.6 mm,5 μm) in methanol-water(V/V=35/65)with the flow rate of 0.8 mL/min and detected at 306 nm in room temperature after injecting 80 μL sample.The linear range of resveratrol was 2~100 μg/mL with an average recovery of 99.1%(RSD=1.2%,n=6).The content of resveratrol in wild and tissue culture of Polygonum cuspidatum were different,and the contents of resveratrol in different parts of Polygonum cuspidatum also varied.The highest resveratrol in Polygonum cuspidatum was observed in wild plants.Though resveratrol of Polygonum cuspidatum differed in different culture conditions,the highest resveratrol content was detected in the roots of wild Polygonum cuspidatum,which indicated that the medical value of wild Polygonum cuspidatum was irreplaceable.
