Cellulose production from fungal mixed culture of Trichoderma reesei RUT C-30 and Apsergillus niger NL02 on steam-exploded corn stover and characteristics of hydrolysis

Abstract
A new approach to cellulase production utilizing steam-exploded corn stover was studied,and it was the mixed culture of Trichoderma reesei and Apsergillus niger to find the balance point of the two strains,spore concentration of A.niger suspensions and spore activation time of A.niger suspensions.The present work used the time courses of cellulase production by monoculture of T.reesei or A.niger as controls.It was found that the highest FPA(filter paper activity) 3.32 U/mL was obtained when the spore concentration of A.niger suspensions was 10 mL-1 and the spore activation time of A.niger suspensions was 12 h,thus it was higher than the FPA of the monoculture of T.reesei,2.25 U/mL.The β-GA(β-glucosidase activity) increased from 0.57 U/mL of the monoculture of T.reesei to 1.32 U/mL of the mixed culture aforementioned.By using enzyme broth from the mixed T.reesei.When 100 g/L steam exploded residue was treated with the enzyme dosage of 20 U/g,the enzymatic culture and the pure culture hydrolysis,the enzyme from mixed culture was 70.00% for 48h,it was higher than the enzymatic hydrolysis from T.reesei of 63.05%.This study demonstrated that mixed cultures of T.reesei and A.niger were feasible and even more optimal than the monoculture of T.reesei or A.niger.
