DETERMINATION OF FERULIC ACID,VANILLIC ACID AND VANILLIN SIMULTANEOUSLY BY TLC IN BIOTRANSFORM PROCESS

Abstract

The contents of ferulic acid,vanillic acid and vanillin in fermentation liquid can be determined simultaneously by TLC,with nhexanechloroformdehydrated etherglacial acetic acid (4∶3∶2∶01) as devekoper and iodine vapor as colordeveloping agent.Vanillin can be determined with 2,4nitrophenyhydrazine as colordeveloping agent specially.The linear range of feruli acid,vanillic acid and vanillin are all between 1~10 μg,their standard curve are y=3533 x-1832,R2=0995 4;y=3080 x-420,R2=0997 9;y=65511 x-490,R2=0995 4 respectively.The average recovery are 1013%,1018% and 1030% respectively,RSD are 239% and 224%.The method of using 2,4nitrophenyhydrazine is more sensitive.The linear range is 1~5 μg,and the standard curve is y=6 8436 x-477 6,R2=0992 2.The determination result of vanillin is same by these two methods.
