THE APPLICATION OF THREE-COLOR FLOW CYTOMETRY IN IMMUNOPHENOTYPING OF LEUKEMIA

Abstract

Objective:To establish a accurate and reliable method in examining immunophenotypes of leukemia.Methods:To use mono color and three color fluorescent staining methods by flow cytometry.Results:Of the 48 cases detected by mono color method,nine cases (18.7 %) couldn't be diagnosed definitely and two cases (4 %) were inconsistent with the final diagnosis.For the other 26 cases detected by using three color method,however,none couldn't be diagnosed definitely or were inconsistent with the ultimate diagnosis.Meanwhile,co expression of related antigens detected by two dimension maps was diagnosed mixed leukemia.Conclusion:As a accurate and reliable method,three color fluorescent staining method can overcome the difficulties in cyto grouping encountered by mono color method and it is extremely important to diagnosis,development and prognosis of leukemia.
