Abstract
An HPLC method with gradient elution mode is developed for the simultaneous separation and determination of chlorogenic acid, caffeic acid,3,5-O-dicaffeoylquinic acid,3,4-O-dicaffeoylquinic acid and 4,5-O-dica-ffeoylquinic acid in flos lonicerae. These five organic acids can be extracted from the flos lonicerae samples with 80% of methanol.Experiments were performed on waters symmetry○R RP18 column(3.9×250 mm,5 μm),with methanol and acetic acid solution(0.1%) as mobile phase using gradient elution at a flow-rate of 0.8 mL/min and detected with a photodiode array detector. The recoveries of five organic acids are 97%~101%,and the relative standard deviations are 0.3%~2.4%.This method can be applied to the determination of five organic acids in flos lonicerae with satisfactory results.
