Abstract
Objective:To predict the transcription initiation site,promoter and transcriptional factors binding sites of claudin-10 gene.Methods:BLAST comparative analysis was used to analyze TATA-box,GC-box and CAAT-box.On-line analysis software Neural Network Promoter Prediction,PROMOTER SCAN and PROMOTER 2.0 were used to analyze the promoter regions on claudin-10 gene 5′UTR,on-line analysis software EMBOSS and CpG Island Searcher were used to analyze the CpG islands,and the on-line analysis software TFSEARCH was used to analyze the transcriptional factors binding sites on claudin-10 gene 5′UTR.Results:One CAAT-box,3 GC-box,4 promoters,2 CpG Islands but no TATA-box was found in Claudin-10 gene.There might be 4 promoter regions,and the CpG islands located between 1700-2143bp or 1367-2200bp,and there were 48 potential promoters in this region.By bioinformatics tool,159 transcriptional factors binding sites were predicted with scores over 85,and 48 transcriptional factors binding sites were predicted when scores were higher than 90,and 9 transcriptional factors binding sites were predicted with scores higher than 95.Conclusion:The prediction of transcription initiation site,promoter and transcriptional factors binding sites in claudin-10 gene is meaningful,though it still needs to be confirmed by further study.
