Study on Isolation of Genomic DNA from the Genus Quercus L. Plants

Abstract

In order to obtain high quality genomic DNA,comparison of the methods for isolating genomic DNA from the genus Quercus L.plants was studied.The results showed that both modified SDS(sodium dodecy sulphate) and CTAB(cetyl triethl am-monium bromide) methods for isolating genomic DNA from the genus Quercus L.plants were better than the original SDS and CTAB methods.DNA yield of modified CTAB method was higher than that of modified SDS method.The isolated DNA was exam-ined by ultraviolet absorption test and agarose gelelectrophoresis.The results showed that the modified CTAB method was suitable for DNA isolation in the genus Quercus L.plants.The purity of the isolated DNA was very high with OD260/OD280 value of 1.75~ 1.80.The genus Quercus L.plants genomic DNA isolated by the modified CTAB method could meet a demand for study of subse-quent molecular such as SSR(simple sequence repeats) and RAPD(randomly amplified polymorphic DNA) analysis.The protein,polyhexose and RNA were eliminated completely.
