Abstract

[Objective] To establish a liquid chromatography tandem mass spectrometry(LC-MS/MS) method for determination of domperidone in human plasma. [Methods] The assay was conducted with an API 3000 LC-MS /MS system consisted of a Ultimate C18 column(50×2 mm,5μm) . The mobile phase consisted of methanol︰water︰formic acid(80︰20︰0.1) and the flow rate was 0.3 ml·min-1. The alkalinized sample was extracted by dichloromethane. After evaporation of the organic layer,the residue was dissolved in mobile phase and the drug was determined by LC-MS.MS using electrospray ionization. Domperidone was detected on multiple reaction monitoring mode by the transitions from the precursor to the product ion(m /z426.4 /175.1) . [Results] Calibration curve was linear in range from 0.125 to 40 ng /ml with a correlation of 0.9995. The intraand inter-day precision(RSD) were all within 2.95%~4.26% and 2.67%~5.00%,respectively,and the accuracy were all within 96.4%-110.6%. [Conclusion] The LC-MS /MS method for the quantitative determination of domperidone in human plasma presented simple and fast sample process,sensitive detection and acceptable precision. Therefore it can be used in determination of the drug concentration in clinic.
