Abstract
Objective To develop a liquid chromatography-tandem mass spectrometry method to determine domperidone in human plasma.Methods After extraction with ethyl ether,domperidone and internal standard were chromatographically separated on an ultimate C18 column(2.1 mm×150 mm,3 μm) with a mobile phase containing 0.1% formate acid buffer and acetonitrile(30∶70,v/v) at 0.25 mL·min-1.The electronic spray ion tandem mass spectrum with the positive mode and multitude reaction monitor(MRM) were used to detect the analytes.Results The linear range of the calibration curves was 0.2-150 ng·mL-1 for domperidone.Intra-day and inter-day RSD was less than 15%.The methodology recovery ranged 99.8%-105.6%.Conclusion The present method is sensitive,efficient and reliable.The described method has been successfully used for the pharmacokinetic studies of domperidone after oral administration in healthy Chinese volunteers.
