Effect of Danshao Huaxian Capsule adding apple on tsp-1,TGF-β_1 of fibrosis liver of rats

Objective: To investigate the effects of Danshao Huaxian Capsule adding apple on the fibrosis liver of rats.Methods: All the rats excepting in the normal control group were subcutaneously given 40%CCl4 injection,high-fat diet(79.5% maizena +0.5% cholesterol +20%lard) and 70%alcohol(every other day) to induce hepatic fibrosis model for eight weeks.Then rats were randomly divided into three groups.After eight weeks,the expressions of TGF-β1 was detected respectively by immunohistochemistry.And the expression of tsp-1 mRNA was determined by Real Time Quantitive RT-PCR.Results: The results from immunohistochemistry demonstrated that: the expression of TGF-β1 was increased in the hepatic fibrosis model and spontaneous recovery group as compared with the normal control group(P0.01);while the levels of TGF-β1 in the Danshao Huaxian group and apple adding Danshao Huaxian group were decreased as compared with the hepatic fibrosis model and spontaneous recovery group.The results from Real Time Quantitive RT-PCR showed that: the expression of tsp-1 mRNA in the Danshao Huaxian group and apple adding Danshao Huaxian group were decreased as compared with the hepatic fibrosis model and spontaneous recovery group(P0.01).While the expression of tsp-1 mRNA in the apple adding Danshao Huaxian group was significantly decreased(P0.01).Conclusion: Apple adding Danshao Huaxian maybe play a better effect in preventing the development of hepatic fibrosis through down regulation the level of tsp-1 mRNA,TGF-β1.
