An Experimental Study on Inducement and Immunological Features of Dendritic Cells from Human Peripheral Blood

Abstract

To amplify dendritic cells (DC) from human peripheral blood and assay their morphological and immumlogical features.The mononuclear cells isolated from human peripheral blood were cultured with the supplement of rhGM-CSF and rhIL-4 to generate DC.The morphological characteristics of those cells were observed and the phenotype features were analysed.The nucleio-type was analysed and the proliferation of allogeneic T lymphocytes stimulated by DC was detected also.Monocytes from human peripheral blood were cultured with rhGM-CSF and rhIL-4 for one week, and then a large number of maturative DC with high puring(90%) were generated,which highly expressed differential antigens CD86, CD40, HLA-DR, CDIa, CD86 and CD83. Meanwhile, the induced DC can effectively stimulate proliferation of allogeneic T lymphocytes.[Conclusion]A large number of maturative DC were induced from human peripheral blood, and may provide foundation for further studies on DC and their clinical applications.
