Abstract
Recombinant plasmid IFN-pPICZα was constructed by fusing porcine interferon-α gene with secretive expression vector pPICZαA.P.pastoris KM71H strain was transformed with linearized IFN-pPICZα.Highly resistant recombinant P.pastoris KM71Hstrains were selected by increasing concentrations of Zeocin~TM .The resistant strains were analyzed to check the presence of inserted genes.Recombinant strain were induced to express PoIFN-α with methanol.Results of SDS-PAGE and Western Blot showed successful expression of recombinant PoIFN-α.WISH/VSV assay showed that the expression product had high antiviral activity(4.1×104 IU/mL).
