Fingerprinting cluter analysis of plants of dendrobium by RAPD

Abstract

Objective To Set up the fingerprinting of species of Dendrobium and discuss the identification of Dendrobium by random amplified polymorphic DNA(RAPD).Methods The RAPD fringerprinting of Dendrobium and its adulterant specise were set up,DNA fragments of RAPD fringerprinting were analysised.Results Eight valid primers produced 53 DNA fragments ranging between 0.25kb and 1.3kb,39 of these fragments were polymorphic bands and accounted for 73.6℅ ;the genetic distance of the twelve Dendrobium spcies varied from 0.04762 to 0.68421,and averaging at 0.44380 which are studied by the cluter analysis;the average genetic distance of Pholidota chinensis and Dendrobium was 0.71734.Conclusions The genetic variations of different species of Dendrobium were rich,RAPD technique was effective in characterizing species of Dendrobium and its adulterant specise.
