OBJECTIVE To investigate the neuroproteelive effect of acetylpuerarm (compound N-2211)and peurarm on focal cerebral is-chemia-reperfusion injury. METHODS The NO level and NOS activity in brain tissue and serum were measured in the rats with reversible middle cerebral artery occlusion( MCAO) without cramectomy. RESULTS The levels of NO and NOS in brain homogenate increased significantly in ischemia-reperfusion group,compared with sham group, and there was the same change happened for the level of NO in serum.NO and NOS levels decreased in acetylpuerarin-treated groups and peurarin-trealed group, compared with ischemia-reperfusion rats. At the same time, the number of living pyramidal cells in CA1 region of hippoeampus increased significantly in acetylpuerarin-treated rats and peurarin-treated rats, compared with ischemia-reperfusion rats. CONCLUSION The results suggested that the effect of acetylpuerarin and peurarin on decreasing NO production played a role on the amelioration of focal brain ischemia-reperfusion injury.
