Abstract
Objective:To evaluate the effect of different doses of Dexamethasome on alkaline phosphatase activity and proliferation of Human dental follicle cells(DFCs) in vitro.Method:Human dental follicle cells of the fifth passage were cultured in DMEM with dexamethasone(0、10~(-10)、10~(-9)、10~(-8)、10~(-7)、10~(-6)mol/ L) and 50ml/L fetal bovine serum.The changes of MTT and the activity of alkaline phosphatase(ALP) were analyzed.Result:Compared with the control group,High-dose(10~(-8)、10~(-7)、10~(-6)mol/L)Dex can inhibit cell proliferation,but low-dose(10~(-10)、10~(-9)mol/L)Dex can't.The ALP activity of human DFCs incubated with Dex(10~(-9)、10~(-8)、10~(-7)、10~(-6)mol/ L)increased significantly.Conclusion:Dex can increase the ALP activity of human DFCs in dose dependent manner and high-dose Dex decrease cell proliferation in vitro.
