Abstract
Objective To explore the inhibitory effect of vasculer endothelial growth foctor(VEGF) antisense oligonucleotides(ASODN) on tumor growth and metastasis of mice Lewis lung cancer.Methods Twenty four mice Lewis lung cancer models were established and subsequently were randomized into three groups: control group,ASODN group,VEGF mismatch sense oligonucleotides(MSODN) group.Sodium chloride,VEGF-ASODN,MSODN were given respectively(once every two days,15 times altogether).The volume and weight of subcutaneous tumors were measured,the rates of lung metastasis were detected by HE staining,while the microvessel density(MVD) in tumor mass was detected by imuunohistochemistry staining,and VEGF protein level in tumor tissue was detected by Western blot assay.Results Tumor growth inhibitory rates of ASODN group and MSODN group were(47.34%) and(7.28%) respectively(P(0.01)).The rate of lung metastasis of ASODN group((37.5%)) was lower than that of MSODN group((75.0%)) and control group((87.5%)).Tumor MVD of ASODN group((12.29)±(2.06)) was lower than that of MSODN group((17.38)±(3.24))and control group((20.14)±(3.90))(P(0.05)).Tumor VEGF protein expression in ASODN group was lower too.Conclusions VEGF-ASODN can inhibit tumor growth and metastasis of mice Lewis lung cancer.So it may become a novel treatment approach for human lung cancer.
