Abstract
Objective To study the expression of IL 1RⅠ protein and mRNA in ischemic rat brain. Methods The expression of IL 1RⅠ protein and mRNA in focal middle cerebral artery occlusion(MCAO)ischemia/reperfusion rat were detected by immunohistochemistry and in situ hybridization. Results Normal and sham rats showed an expression of IL 1RⅠ protein and mRNA mainly in cortex. The number of IL 1RⅠ protein positive cells increased after ischemia. It increased by 6 hours and reached the peak at 12 hours in ischemic cortex after reperfusion. The cells express IL 1RⅠ mRNA were increased after 2 hours of reperfusion, maximally increased in cortex by 6 hours of reperfusion and prolonged up to 24 hours of reperfusion. Conclusion Our results indicate that the upregulated expression of IL 1RⅠ could bind IL 1β competing with IL 1RⅡ and IL 1 receptor antagonist, then transduct the signal of IL 1β enhancing the ischemic damage.
