Abstract
Objective To observe the effects of spironolactone on left ventricular function and collagen remodeling after actue myocardial infarction(MI).Methods AMI was induced by ligating the left anterior descending coronary artery in rats.24 hours after coronary ligating,the rats were randomly divided into four groups and treated for six weeks.(1)MI+placebo;(2)MI+small-dose spironolactone(SSpi);(3)MI+high-dose spironolactone(LSpi);(4)sham-operation group.The Mean blood press(MAP),left ventricular end-diastolic press(LVEDP),interstitial collagen content and the ratio of type Ⅰand Ⅲ collagen in non-infarction area were detected at the 2nd week and the 6th week.Results As compared with that in sham-operation group, LVEDP in placebo-treated infracted rats was increasedsingnificantly(P0.01),so did the collagen content(P0.01) and the ratio of type Ⅰand Ⅲ collagen,as compared with that of sham group at the 2nd week and the 6th week(P0.01).In spironolactone treated rats the collagen content,the ratio of type Ⅰand Ⅲ collagen ,MBP and LVEDP were significantly decreased,as compared with those in placebo-treated rats(P0.05 or P0.01),however, L-Spi treated rats were lower in collagen content and the ratio of type Ⅰand Ⅲ collagen at the 2nd week and the 6th week,and LVEDP at the 6nd week than S-Spi treated rats (P0.05).Conclusion Spironolactone couldimprove the left ventricular function and has beneficial effects on the left ventricular collagen remodeling after myocardialinfarction.
