Abstract
Objective To observe the effects of spironolactone and losartan interventions on homodynamics and collagen remodeling after myocardial infarction.Methods During 2005.10 to 2006.3,we have done the study at Hebei Academy of Medical Sciences.The myocardial infarction model was induced by ligating the left anterior descending coronary artery.Then the rats were randomly divided into sham,placebo,spironolactone,losartan and losartan+spironolactone groups.Mean blood pressure(MBP),left ventricular end-diastolic pressure(LVEDP),interstitial collagen content and the ratio of type Ⅰand Ⅲ collagen of non-infarction zone were assessed at the 2nd and the 6th week.Results Significant increase in LVEDP,collagen content,and the ratio of type Ⅰand Ⅲ collagen(P0.05)were found in the placebo-treated infracted rats as compared with sham group at the 2nd and the 6th week.MBP,LVEDP and the ratio of type Ⅰand Ⅲ collagen contents were all significantly decreased in lasartan treated group and the combination treated group than placebo-treated rats at the 2nd week.At the 6th week,LVEDP,collagen contents,the ratio of type Ⅰand Ⅲ collagen were all significantly decreased in spironolactone treated rats,losartan treated rats and the combination treated rats.However,MBP,LVEDP,collagen contents,the ratio of type Ⅰand Ⅲ collagen were lower in the combination treated rats compared with spironolactone treated rats and lasartan treated rats.Conclusion Spironolactone and losartan can coordinate and improve the cardiac hemodynamics and the left ventricular collagen remodeling after myocardial infarction in rats.
