Study on apoptosis of peripheral neutrophil and Mcl-1,caspase-3 expression in preeclamptic patients

Abstract
Objective:To investigate peripheral neutrophil apoptosis in preeclampsia patients and its mechanism.Methods:Neutrophils isolated from 20 preeclamptic patients and 20 normal pregnancies were incubated with autologous serum in vitro for 24 h.Then the apoptosis rate was determined by Flow Cytometry,and Mcl-1 mRNA expression was determined by RT-PCR,caspase-3 protein expression was determined by Western blotting.Results:Apoptosis rate was significantly reduced in neutrophils from preeclamptic patients(16.51%±3.84%) compared with that of normal pregnancies(30.62%±6.27%).In preeclamptic group,Mcl-1 mRNA expression elevated,but caspase-3 protein expression decreased.Conclusion:Apoptosis in peripheral neutrophil of preeclampsia delays,the upregulation of Mcl-1 and downregulation of caspase-3 may act an important role in the apotosis delay.
