Study on the Expression of Hypoxia-inducible Factor-1α,VEGF and sFlt-1 in Preeclamptic Placenta

Abstract

0bjective: To investigate the expression and correlation of HIF-1α and VEGF in the preeclamptic placenta,aiming to discuss the significance in the pathogenesis of preeclampsia.Methods:Placenta were collected from 20 pregnant women with preeclampsia as study group and 15 normal pregnant women as control group,the expression of HIF-1α and VEGF protein were semi-quantitatively analyzed with immunohistochemicall assay and mRNA level was determined by using reverse transcription polymerase chain reaction(RT-PCR) technique.Results:(1)The expression of HIF-1α protein in preeclamptic group obviously increased,with(+++)positive expression rate significantly higher than control group(P0.05),but VEGF expression obviously reduced in preeclamptic group(P0.01).(2)The level of HIF-1α mRNA in preeclamptic placenta was significantly higher than normal placenta(P0.05),although the level of VEGF mRNA increased in preeclamptic placenta,but not significantly different from normal placenta(P0.05).Conclusion:The abnormal higher HIF-1α expression in preeclamptic placenta indicated that HIF-1α might play an important role in the pathogenesis of preeclampsia,possibly through affecting the cytotrophblastic invasion and placental vascular reconstruction via the modulation of VEGF gene transcription.
