Abstract

Objective To investigate the sensitivity and specificity of DHPLC detection on mutation site of susceptibility genes in breast cancer.Methods 202 patients diagnosed as primary breast cancer were enrolled,PCR/DNA and DHPLC were simultaneously applied to screen the BRCA1 gene mutation.PCR/DNA was used as golden standard,and the sensitivity and specificity of DHPLC on gene mutation detection was analyzed.Results Overall the 202 samples,76 samples were positive for BRCA1 gene mutation detected by PCR/DNA while 73 samples were positive for BRCA1 gene mutation with DHPLC,and the sensitivity of DHPLC was 96.1%.The number of negative samples were 126 with PCR/DNA detection and were 120 with DHPLC detection,and the specificity was 95.2%.There were no statistically significant of sensitivity and specificity of PCR/DNA and DHPLC on the screening of BRCA1 gene mutation.Conclusion The sensitivity and specificity of DHPLC on the detection of BRCA1 gene mutation in breast cancer was well,and compared with the traditional methods,DHPLC was with many virtues and deserved broadly application.
