Abstract
Objective It is to observe the influence of Danxingtongluo decoction(DXTLD) on nerve cell heat shock protein 70(HSP70) expression in rats with cerebral ischemia reperfusion injury and to discuss its nerve protection mechanism.Methods Forty healthy male Wistar rats were chosen to establish focal cerebral ischemia reperfusion models with middle cerebral artery thread embolism method and were randomly divided into sham operation group,ischemia/reperfusion(I/R) group,DXTLD high dosage group,DXTLD low dosage group and Nimodipine(NIMO) group. There were eight rats in every group.DXTLD groups were given DXTLD at doses of 15 mL/kg and 7.5 mL/kg.NIMO group was given NIMO at dose of 10 mg/kg.The drugs were given once a day and consecutively for seven days before operation.Normal saline were given to sham operation group and I/R group at dose of 10 mL/kg.After 2 h ischemia and 24 h reperfusion injury,the rats were killed and their brains were taken out.The HSP70 expression in brain tissue was measured with immunohistochemical method and was compared with the change of pathohistology.Results DXTLD could obviously increase the HSP70 expression in cerebral cortex and hippocampus and relieve nerve cell injury.Conclusion DXTLD can relieve ischemia reperfusion injury of brain tissue and improve nerve function lose.The mechanism may be related with the increasing of HSP70 expression.
