Effects of IL-22 and IL-22 siRNA on Proliferation and Apoptosis of HaCaT Cells

Abstract

Objective To investigate the effects of interleukin-22( IL-22) and IL-22 siRNA transfection on proliferation and apoptosis of HaCaT cells. Methods After being treated with IL-22 in different doses( 12. 5μg / L, 25μg / L,50μg / L,100μg / L,200μg / L) and IL-22 siRNA,proliferation of HaCaT cell was determined by MTT. Apoptosis rate was assessed with flow cytometry. Results Proliferation of HaCaT cell treated with IL- 22 increased significantly when compared with that of the control. Meanwhile,proliferation of HaCaT cell treated with IL-22 siRNA decreased significantly when compared with that of the control. The apoptosis rate of HaCaT cell treated with IL-22 was lower than that of the control,but treated with IL-22 siRNA was higher than that of the control,and the difference were statistically significant. Conclusion IL-22 can promote HaCaT cell proliferation and inhibit apoptosis effectively. The effect is related to the concentration of IL-22. IL-22 siRNA can inhibit HaCaT cell proliferation and promote apoptosis effectively.
