Abstract
Objective To construct and express a mammal expression vector coding human thioredoxin (hTRX) gene. Methods The human thioredoxin cDNA was inserted into pGEM-T Easy vector. A mammal expression vector coding human thioredoxin gene(pShttle-hTRX) had been constructed and transfected into HeLa cells after confirming the sequence. The structure of the recombinant vector had been verified by Western blot, immunohistochemistry, activity examination, restriction analysis and PCR amplification. Results The recombinant mammal expression vector coding thioredoxin gene was constructed, and the transcription and expression of hTRX were detected. Conclusion The recombinant mammal expression vector coding thioredoxin gene has been successfully constructed, which helps to study the biochemical role of thioredoxin.
