Abstract

Objective:To establish an HPLC method for determination chlorogenic acid and 3,5-O-dicaffeoylquinic acid in Ainsliaea fragrans Champ.Method:The chromatographic column was Discovery C18(250 mm×4.6 mm,5 μm),UV detection wavelength was 328 nm,a mixture of 0.05% formic acid-water and acetonitrile was used as mobile phase with gradient elution and flow rate was 1.0 mL·min-1.Results:Chlorogenic acid at the quality of 0.60-3.00 μg showed good linearity,the average recovery(n = 5)was 100.8% ;3,5-O-dicaffeoylquinic acid at the quality of 1.30-6.52 μg showed good linearity,the average recovery(n=5)was 99.4%.Conclusion:This methods is fast,reliable and accurate,and may be as one of index used to judge the quality of Ainsliaea fragrans Champ.
