Abstract

Objective: An HPLC method for the simultaneous determination of Chlorogenic acid and Caffeic acid in Flos Lonicerae was proposed based on rapid ultrasonic-assisted extraction using the non-ionic surfactant TritonX-100 as the extractant. Methods: The separation was performed on Phenomenex C18( 250 mm ×4.6 mm,5 μm) column with detection wavelength of 326 nm. The mobile phase consisted of methanol-0. 6% acetic acid solution( 25 ∶ 75,V /V) with the flow rate of 0. 80 mL /min,and the column temperature was set at 35℃. Result: The calibration curves for Chlorogenic acid and Caffeic acid were liner between 0. 00076 ~ 190. 00 μg /mL( r = 0. 99995),0. 000864 ~ 216. 00 μg /mL( r =0. 99966) respectively. The limit of detection( LOD) were 0. 029 ng /mL,0. 030 ng /mL,respectively. The recoveries of the two compounds range were 95. 13% ~ 100. 24%. Conclusion: The method was simple,rapid,convenient,sensitive,and environment-friendly,and thus can provide new and scientific means for assaying the contents of Chlorogenic acid and Caffeic acid in Flos Lonicerae.
