Research of effect of major herbs in Jianpihuoxue Decoction against acute hepatic injury in rats induced by lipopolysaccharide

Objective: To investigate the major herbs in jianpihuoxue decoction against Lipopolysaccharide injury and find the action point of major herbs.Methods: The rats were injected in tail vein with Lipopolysaccharide to make the acute hepatic injury model.And uniform design was adopted to design the experiments.Portal vein plasma TNF-α was detected as the index.Screen out the major herbs and verify the efficacy with the same model.The protein expression of CD68,CD14,TLR4,P-IκB and the expression of LBP,TLR2,TNF-α,IL-1β,IL-6 mRNA in hepatic tissue were detected.Results: Fructus aurantii,White peony root,Alisma and Schisandra were the major herbs in jianpihuoxue decoction against Lipopolysaccharide injury.The major herbs could reduce the plasma TNF-α significantly.The major herbs could decrease the protein expression of CD68,CD14,TLR4,P-IκB,especially in decreasing CD68 which was better than Jianpihuoxue Decoction;LBP,TLR2,TNF-α,IL-1β,IL-6 mRNA expression in hepatic tissue were reduced apparently also,and the efficience of the major herbs was even better than Jianpihuoxue Decoction.Conclusion: The combination of Fructus aurantii,White peony root,Alisma and Schisandra were the major herbs of jianpihuoxue decoction against acute hepatic injury induced by Lipopolysaccharide.The action point of the major herbs was inhibiting the activation of kupffer cells.
