Effect of Fenofibrate on liver injury with obstructive jaundice

Abstract

【Objective】 To investigate the effect of Fenofibrate on serum liver function,cytokine,hepatocyte apoptosis and collagen fiber with obstructive jaundice.【Methods】 Rats were subjected to bile duct ligation to establish the cholestasis mode1.Forty-eight male Wistar rats were randomly divided into four groups equally: Group A = normal group,Group B = bile duct ligation(BDL) only,Group C = BDL + Fenofibrate [30 mg/(kg·d)],Group D = BDL + Fenofibrate [60 mg/(kg·d)].All rats were sacrificed on 7th day after obtaining blood samples and liver tissue.Serum ALT,AST,ALP,GGT,TBA were tested by Automatic Chemistry Analyzer,serum TNF-α,IL-1βwere tested by radioimmunoassay kits,the number of hepatocyte apoptosis were tested by TUNEL and collagen fiber was tested by Masson.【Results】 Serum ALT,AST,ALP,GGT,TBA levels in rats of group D were significantly lower than in group B(P 0.01),while Serum AST,ALP,GGT,TBA levels of group D were significantly lower than in group C(P 0.01);L-1β level of group D is lower than group C(P 0.01),when IL-1β level of group C and D is lower than group B(P =0.000,0.001),TNF-α of group D is lower than group B(P 0.01);hepatocyte apoptosis in the animals of group C were significantly lower than in group B(P 0.01) when hepatocyte apoptosis in group D were significantly lower than in group B or C(P 0.01);collagen fiber area of group C is smaller than group B(P 0.01),while collagen fiber area of group D is smaller than group B and C(P 0.01).【Conclusion】 Short administration of Fenofibrate can significantly attenuate cholestasis-induced liver injury by improving serum liver function,suppressing inflammatory response,inhabiting hepatocyte apoptosis and performing anti-hepatic fibrosis effect.
