【Objective】To investigate the expression of 5-lipoxygenase in the liver during obstructive jaundice.【Methods】Forty eight male Sprague-Dawley rats were randomly divided into three groups: sham-operation group(sham),bile duct-ligation group(BDL),and bile duct-ligation plus AA-861 group(AA-861).The rats were sacrificed at 7 and 14 days after operation.The levels of plasma endotoxin were measured using limulus test;Immunohistochemitry were used to detect the expression of 5-lipoxygenase of liver.The serum levels of LTB4 and TNF-ɑ,IL-6 were analyzed by enzyme-linked immunosorbent assay(ELISA).To test the content of MDA and SOD of the liver tissue by homogenate,ALT and AST were test by Automatic biochemical analyzer.【Results】Expression of 5-LOX was significantly lower in sham group,the expression of 5-LOX was obviously increased in BDL group,and the expression of 5-lipoxygenase was significantly lower at each time point in AA-861 group than in BDL group.The serum levels of LTB4,TNF-ɑ,IL-6,ALT,AST and endotoxin were significantly lower in sham group than BDL and AA-861 group,The serum levels of LTB4,TNF-ɑ,IL-6,ALT,AST were significantly lower at each time point in AA-861 group than in BDL group,the liver tissue levels of MDA was significantly lower in AA-861 group than in BDL group,the liver tissue levels of SOD was significantly higher in AA-861 group than in BDL.【Conclusion】Expression of 5-LOX is significantly higher during obstructive jaundice,AA-861 may protect against the liver injury in rat with obstructive jaundice through reducing the expression of 5-LOX in liver.
