Abstract

Objective To explore the effect of flavonoids effective components from Astragalus and Kudzu when decocted with different methods on interventing glucose and lipid metabolism in diabetic rats and its related mechanisms.Methods According to random blood glucose,55 SD male rats were divided randomly into 5 groups,such as normal control group,model control group,positive control group( Jin Qi hypoglycemic tablets),Astragalus flavonoids Kudzu flavonoids group( divided decoction group),and total flavonoids from Huangqi Gegen decoction( mixed decoction group).Except the normal control group,the others were injected singlely Streptozocin( STZ) via intraperitoneal to make diabetic models.After administrating 30 days,blood glucose,serum cholesterol( CHO) and triglyceride( TG) were tested,and hepatic insulin( INS),glucose transporter 4( GLUT4),adiponectin( ADPN),and leptin( LEP) were tested through enzyme- linked immunosorbent assay( ELISA) Kits as well.Results Compared with the normal group,the glucose,CHO and TG levels elevated,while the levels of hepatic INS,GLUT4,ADPN and LEP were decended.Both divided decoction group and mixed decoction group could reduce levels of blood glucose,serum TG and CHO in diabetic rats( P 0.05),and there were no differences between the two groups( P 0.05).In regulating on glucose metabolism,divided decoction group could significantly increase INS,GLUT4 levels,while mixed decoction group raised GLUT4 level obviously,and the effect of divided decoction group was better than the mixed one( P 0.05).In regulating on the relevant adipokines,both divided decoction group and the mixed one could higher hepatic LEP level identically( P 0.05),however,both of them had no influence on hepatic ADPN level.Conclusion The flavonoids effective components from Astragalus and Kudzu when decocted separately and as a whole showed no significant differences on the effect of lowering blood glucose and lipid,while in regulating on INS and GULT4 levels,the effect of decoction separately was better.
