Optimization of Two-phase Systems Extraction Total Flavonoids in Phellodendron by Response Surface Analysis

Abstract

Total flavonoids in phellodendron was extracted by alcohol-ammonium sulfate two-phase aqueous system synergism with ultrasound extraction.Extraction time,ethanol-water ratio,material-solvent ratio and salt concentration on extraction ratio of total flavonoids were studied by experiment of single factor.Basing on single factor experiment,extraction time,ethanol-water ratio and material-solvent ratio were as response factors.Extraction ratio of total flavonoids from phellodendron was as response value.According to Box-Benhnken principle of design,the response surface method included three response factors and three levels and influence conditions on extraction ratio of total flavonoids were determined.The results of experiment showed that the maximum technical conditions were:extraction time 77min,material-solvent ratio 1:26(m/V),ethanol-water ratio 0.71(V/V).
