Abstract
In order to select the best way to extract genomic DNA in Mycoplasma for molecular identify and PCR detecting.Using two kit products,Phenol/chloroform/isoamyl alcohol method,CTAB method,high salt method and boiling method extracted genomic DNA from Mycoplasma culture media of two strains,evaluated their useful efficiency as templates in PCR procedure after optimization.The Results indicted: All the genomic DNA samples could be detected by PCR.CTAB method and imported kit yielded the best quality DNA,Phenol/chloroform/isoamyl alcohol method,high salt method and kit made in China produced lower quality DNA,and the worest quality product resulted from boiling method.CTAB method and high salt method could be the optimal ways for obtaining the genomic DNA.
