Abstract

This study aimed to investigate the frequencies of genotype and allele distribution of Toll-like receptor 4(TLR4) gene at 3′-untranslated(3′-UTR) region single nucleotide polymorphism(SNP) in Guangxi population, and to analyze allelic and genotypic diversity of TLR4 gene among different regions and different races. Polymerase chain reaction-single base extension(PCR-SBE) and DNA sequencing methods were used to analyze the polymorphism of rs11536889G/C at the TLR4 gene 3′-UTR region. The genotypic and allelic diversity of rs11536889G/C at the TLR4 gene 3′-UTR region were compared with other population(Hapmap-CEU, HapmapHCB, Hapmap-JPT, Hapmap-YRI) from the Human Genome Project group(Hapmap) data(http://www.ncbi.nlm.nih. gov/projects/SNP/) as well as from Korea. The results showed that the diversity of polymorphisms had no significant difference between female and male groups of Guanxi population(P 0.05); and no significant difference(P 0.05) of genotypic and allelic diversity were observed when compared to Hapmap-JPT, Korea and Hapmap-HCB; while there were significant difference(P 0.05) of genotypic and allelic diversity when compared to Hapmap-CEU and Hapmap-YRI. In conclusion, the frequencies of polymorphisms distribution at the TLR4 gene 3′-UTR region had significant difference between Guangxi populations and other regional and ethnic populations.
