Preliminary Studies on Apoptosis in Human Osteosarcoma Cells Induced by Cisplatin and Its Molecular Mechanism

Abstract

Objective To investigate the possibility and the mechanism of human osteosarcoma cells apoptosis by cisplatin(cDDP).Methods After treatment with cDDP,the cytotoxic activity of cDDP was quantified by determining IC 50 in human osteosarcoma cells using trypan blue exclusion,and apoptotic cells were detected with flow cytometry,DNA cytofluorometry,TUNEL and DNA electrophoresis.Results cDDP inhibited the growth of human osteosarcoma cells in a dose-dependent manner in a certain range with a IC 50being 1.05 μg/ml±0.25 μg/ml;Characteristic morphologic peak of apoptosis and DNA ladder were observed in human osteosarcoma cells treated with 0.5~2.0 μg/ml.Conclusions Could induce apoptosis of human osteosarcoma cells in vitro,which should be the basis of its effectiveness.cDDP showed great poential in the treatment of osteosarcoma.
