Effect of Lupus Fang I on Preventing Hydrocortisone Damage to Chondrocyte

Abstract

Objective To observe the effects of lupus FangⅠon preventing hydrocortisone damage to chondrocyte. Methods The chondrocyte was isolated from cartilage of 3~4 weeks newly born rabbit by means of collagenase digestion and identified by image analysis, toluidine blue staining, alcian toluidine blue staining, etc. These chondrocyte systems were divided into hydrocortisone groups, traditional Chinese and western medicine groups, traditional Chinese medicine groups and blank groups at random. Different groups of the mixed liquor of lupus FangⅠand hydrocortisone were incubated for 12 hours and 36 hours. Morphologic changes of chondrocyte were observed through inverted microscope. The effect of the mixed liquor of lupus FangⅠand hydrocortisone on the proliferation of chondrocyte was monitored by MTT analysis. Results From observing morphologic change of chondrocyte, chondrocyte were died or major injured in hydrocortisone groups, and the chondrocyte growed well in traditional Chinese and western medicine groups, traditional Chinese medicine groups and blank groups. The results of MTT analysis were showing no difference with chondrocyte amorphous observation. Conclusion Lupus FangⅠhas effect on preventing hydrocortisone damage to chondrocyte.
