Effect of Osteoarthritis FormulaI Against Chondrocyte Lesion Induced by Hydrocortisone in Rabbits

Objective:To observe the effect of osteoarthritis formulaⅠ against chondrocyte lesion induced by hydrocortisone . Method:The chondrocyte was isolated from cartilage of 3～4 weeks newly born rabbit by means of collagenaseⅡdigestion and identified by staining method of alcian bllie and toluidine blue. Different doses of the mixed liquor of osteoarthritis formulaⅠand hydrocortisone were added to the chondrocyte and incubated . Morphologic changes of chondrocyte were observed through inverted microscope,and the effects of the mixed liquor of osteoarthritis formulaⅠ and hydrocortisone on the proliferation of chondrocyte were monitored by MTT analysis. Result: 1.25 ,2.5 mg/ml hydrocortisone almost made chondrocyte die on cultivation in vitro for 24 h, meanwhile the cell grew well when both the mixed liquor of 1.25 mg/ml hydrocortisone and 150 mg/ml osteoarthritis formulaⅠand the mixed liquor of 2.5 mg/ml hydrocortisone and 300 mg/ml osteoarthritis formulaⅠwere added to the chondrocyte and incubated, respectively. The result of MTT analysis accorded with cartilage cell morphologic observation. Conclusion: Osteoarthritis formulaⅠcould prevent against chondrocyte lesion induced by hydrocortisone.
