Abstract
Objective Beta defensins are small antimicrobial cationic peptides.Beta-defensin-2 is inducible expressed throughout the epithelium and the serous gland cells.This article aims to construct the prokaryotic expression vector of human beta-defensin-2 gene and establish foundation of humanβ defensin-2(HBD-2)gene expression,purification and production for gene engineering.Methods Gene fragment of humanβ defensin-2(HBD-2)gene was composed by exopathic gene and cloned into pMD18T.After identified by enzyme digestion,it was cloned again and inserted into expressing plasmid pET32a,then,it formed fusion gene with TrX-A from vector.Results Plasmid pET32a-HBD2 was constructed,identified by analysis,and it didn't appear gene mutation.Conclusion pET32a-HBD2 has been constructed successfully.
