Abstract
Objective To clone and construct eukaryotic expression vector of human beta-defensin 3.Method Full length cDNA of human beta-defensin 3(hBD3) was cloned by RT-PCR technique from human skin and then inserted into pcDNA3 vector followed by transformation to DH5α competent cell. After restriction analysis, the recombinant vector was further identified by sequencing.Result The DNA fragment of interest was proved to be identical to human beta-defensin 3; The full-length cDNA in recombinant pcDNA3-hBD3 was demonstrated correct, without any mutation.Conclusion We successfully cloned the full-length cDNA of human beta-defensin 3 and constructed its recombinant eukaryotic expression vector pcDNA3-hBD3.
