Abstract
With the aim to improve the purity of chlorogenic acid(CA), the suitable resin for separating and purifying CA of Lonicera japonica were studied, and purifi cation were achieved by column chromatography with the material of crude extract. The characteristics of static adsorption dynamics and adsorption isotherm of macroporous resins to CA were investigated. The effect of initial p H on adsorption and the concentration of elution on desorption were also investigated. The parameters of purification were determined. The results indicated that the ADS-21 macroporous resin gave the best effect of separation with the initial p H 2.0 of crude extract, the sampling rate of 0.6 m L/min, the long to diameter ratio of 25︰1, 70% ethanol solution eluting at the rate of 0.6 m L/min. Under the conditions, the purity of product with 52.08% is obtained, the yield can reach 89.49%.
