Abstract

[Objective] The paper aimed to study on the optimum extraction and purification process of Chlorogenic Acid from Lonicera japonica Thunb.[Method]Preliminary test with 70% ethanol and water were used to determine the extraction solvents,the optimum condition of extraction process were extracted by the orthogonal design.In the purification experiment obtained optimum scheme by the ethyl acetate extraction and the polyamide column chromatography.The chromatography conditions of the polyamide chromatography and the elution effect of different elution solvent were observed.[Result]The optimum extraction condition of chlorogenic acid in L.japonica Thunb were: with 70% concentration ethanol as extract solutions,adding alcohol amount 12 times,refluxing for 3 times,2 h each time.Average extract rate was 6.62%.In the ethanol acetate extraction,the purity of the chlorogenic acid was 18.4%,the 30% concentration ethanol was the best as elution solvent,the purity of chlorogenic acid was 33.2%.[Conclusion]Compared with organic solvent purified method,polyamide chromatography had advantages of simple operation,high purity,no residual organic solvents and low cost.
