Abstract

Objective To establish the hepatocyte steatosis models with human hepatocyte strain L-02.Methods The growing cells L-02 were treated by different concentrations of oleic acid.To screen the proper concentration of oleic acid,the proliferation of cells were measured by MTT.Lipid droplets in the cells were observed through oil red staining.The levels of TG,ALT and AST were also measured with analyzed kits.Results Lipid droplets were observed through light microscope and electron microscope by treating the cells with 20μg/ml oleic acid for 3d.Compared with the control group,the level of TG,ALT and AST in model group also increased(P0.01).Conclusion The steatosis models of the hepatocytes were established successfully by treating with oleic acid.
